Let the no job things run for longer, and once the building becomes abandoned, it will tell you if it's long commute time or low desirability. I don't know the size of your city, but getting jobs from other tiles have always been flaky for me. My sims generally can't travel more than 1 large tile in distance for jobs.

Not so sure that "letting the no jobs thing run for longer" and queing the buildings after abandonment is the best option.  To find out if it's low desirability, all you need to do is check the desiribality data on your dashboard (green=high desirirability, red=poor desiribality).

Also, the "abandoned due to lack of commute" can be a rather vague message, as it will pop up even if it's not because of a long commute, but because the game doesn't really know why the building was abandoned...

Do your neighboring cities have a lot of people?  If so they could be taking the jobs there and not leaving any for your problem city...I would try to zone/grow commercial or industrial (whatever is in higher demand) right in this city so you can provide jobs for these buildings.

Keep calm and take photographs.

If it was working fine, I expect you did something that broke the road connections to work.. all I can think of.. yoir city is so small (220) that there are not a lot of things that can happen that early.. use the "route query" tool and look for places where and why the R's can't get to work.. You can also try to hit "cntrl-x" and enter "drawpaths" in the window and hit return (without the quotes" and it will draw arrows where things can move on your road, rails, etc.. look for places where they arrows are "broken"

Hmm... That's interesting. I'm assuming you didn't delete the neighbor cities (hopefully you didn't!). Are the connections to the neighboring city close enough for the residents to find the jobs attractive? Because if these 220 people have to go across one large tile to get to jobs, they aren't gonna go. (Well, maybe the crazy ones). A picture would definatly be helpful.

Make sure they're close enough. Also do make sure there is no broken road connection. I had a similar issue except I noticed my highway connection wasn't completed with onramps on both sides. Without em, my sims I guess would get on the highway and never get off thus never reaching their job. Otherwise, try demolishing your existing connection, then reestablishing the connection.
