Is it better for me to build multiple cities in a region and connect them rather than trying to fit it all on one tile?

Yes, for a variety of reasons. Remember, demand is shared over all connected cities in a region, as are stage caps.

It is best to build multiple regions since you can put undesirable items (such as landfills, power plants, industry) in one tile and then the rest in another. As His Divine Hand said, demand is spread across cities so don't worry about anything like that. Also, it adds a realistic type of touch to your cities. Seeing freight trucks driving out into a blank tile is lame, but knowing they're actually going somewhere is nice. Also, it allows of better city planning and encourages city growth. Single cities can easily become stagnant at around 5-10k pop. By building multiple cities, you allow that city to grow! hope this helps!
