City Sizes and Shapes

That's not possible there are only 3 city sizes. small(red) medium (green) Large (blue).

unfortunately, petertoer is correct - it is impossible to make the larger cities even larger, and it is impossible to change the shapes of the cities - they will forever remain square.

You could create a non square shaped region though.  Then simply turn off the borders and it would look pretty cool

Yes I think you can do that on the menu in region view.

To make a non square region, just put black in the config map where you don't want the city tiles to be. Black won't show up as a city in region view. When people say you can make an any shape and size region they mean you can make the region any squared shape you want. You can't make a circular region. The only thing that determines how big the size of your region is, is your computer specs.
